During the past decades, one of the main challenges in chemical synthesis has been to fulfill the demand of enantiopure compounds from the pharmaceutical industry, but also of other major sectors including the fragrance, agrochemical, food, and materials industries. A major obstacle to the development of such compounds is the difficulty associated with synthesizing large quantities in an economical fashion, often because complex multi-step synthesis is usually required. The major goal of this proposal, ChiralCreator, is to develop new asymmetric activation modes and their application to simplify the synthesis of complex enantioenriched molecules. We will exploit
photoredox catalysis for the generation of new radical intermediates that, upon their coordination with a chiral metal catalyst, will evolve through unconventional asymmetric C=C and CH bond activations. ChiralCreator will provide new disconnection approaches using abundant feedstocks, for the synthesis of current enantioenriched medicines as well as analogues that are difficult to synthesized using current methodologies.
